摘要：afullerene−paclitaxelconjugatehasbeensynthesizedasaslow-releasedrugforaerosolliposomedeliveryofpaclitaxelforlungcancertherapy.theconjugatewasdesignedtoreleasepaclitaxelviaenzymatichydrolysisandsubsequentlyhasshownahalf-lifeofreleaseof80mininbovineplasma.aliposomeformulationoftheconjugatehasbeenpreparedusingdilauroylphosphatidylcholine(dlpc),anditsic50isvirtuallyidenticaltotheic50forapaclitaxel−dlpcformulationinhumanepitheliallungcarcinomaa549cells.withbothclinicallyrelevantkineticsofhydrolysisandsignificantcytotoxicityintissueculture,theconjugateholdspromiseforenhancedtherapeuticefficacyofpaclitaxelinvivo.
